The modern pharmaceutical industry is facing unprecedented challenges in drug development. In spite of the doubling of global research funding since the beginning of the last decade, the sequencing of the human genome and the introduction of technologies such as combinational chemistry and high-throughput screening, the number of new chemical entities approved by the FDA has continued to fall over the past ten years.

The strategy adopted to date in drug development has been to focus on a single target asociated with a particular pathological mechanism. However, many human diseases are highly variable and heterogeneous, involving multiple organ systems, tissues and potential targets and these cannot be addressed by single-drug therapies targeted to a specific mechanism or receptor site.

Xenexus Pharmaceuticals is developing single compound multifunctional drugs that simultaneously target a number of different pathological mechanisms that are associated with complex diseases such as multiple sclerosis, stroke, rheumatoid arthritis and Alzheimer’s disease. These drugs are already marketed for other indications which means that the potential benefits of multifunctionality can be added to the advantages of drug reprofiling such as known pharmacokinetics, safety in clinical use and decreased time in development.
